  Université de Montréal.\nRésumé :Genome-wide CRISPR/Cas9 knockout sc
 reens can reveal genotype-to-phenotype relationships and mechanisms of res
 istance and sensitization to drugs and other compounds. We have undertaken
  more than 250 CRISPR/Cas9 screens to map these relationships. I will pre
 sent a plan to apply network-based approaches to model fitness effects of 
 gene knockouts to predict drug mechanisms\, gene functions\, cancer-specif
 ic vulnerabilities and synergistic drug combinations.  I will also cover 
 earlier work on the network-level effects of alternative splicing and dise
 ase variants and present a machine learning-based strategy to reconstruct 
 signaling pathways mediated by short linear motifs in disordered protein r
 egions.
