SUMMARY:In vivo metabolic rewiring via CRISPR-Cas9-based genome editing
DESCRIPTION:Conférence scientifique | Centre de recherche du CHU Sainte-Ju
 stine
\nConférencier :Yannick Doyon\, PhD (invitée pour l'axe Santé m
 étabolique et cardiovasculaire)
\n
\nChercheur régulier\, Axe Reproducti
on\, santé de la mère et de l’enfant\, Centre de recherche du CHU de Q
 uébec – Université Laval
\nProfesseur adjoint\, Département de médec
 ine moléculaire\, Université Laval
\n
\nBiographie :I established my lab
oratory at the CHU de Québec–Université Laval research center in Septe
mber 2013 after a seven-year postdoctoral stint in the biotechnology secto
r (Sangamo Therapeutics\, California\, USA) devoted to the creation and im
plementation of genome-editing tools. My team develops versatile methods f
or increasing the efficacy of targeted genome editing to enable the creati
on of bona fide cellular models for biological research. In addition\, we
explore the potential of in vivo genome editing as a novel class of human
 therapeutics for pediatric metabolic disorders.
\nRésumé :In this semina
r\, I will share our recent results demonstrating that CRISPR-Cas9 systems
can be used to dissect metabolic pathways in vivo. Using a mouse model of
hereditary tyrosinemia\, we uncovered intra-pathway enhancers and suppres
sors that modulate the phenotype and the levels of key catabolites of the
pathway. I will also describe our approach to streamline the incorporation
of marker-free genetic changes in human cells (Agudelo et al\, Nature met
 hods 2017).
