\nRÉSUMÉ: Ipatasertib (GDC-0068) is a small-molecule Akt kinase i
 nhibitor in late-stage clinical trials for oncology. We will describe our 
 research and development efforts aimed at defining an efficient process to
  synthesize this complex active pharmaceutical ingredient. We will illustr
 ate the interplay of process chemistry\, organometallic catalysis\, biocat
 alysis and API solid-state final form control considerations in the evolut
 ion towards an efficient route that bears the hallmarks of a commercial ma
 nufacturing process.
